Research Finding Could Lead to Targeted Therapies for Inflammatory Bowel Diseases
Researchers have shown that a pathogen-sensing molecule plays a vital role in keeping gastrointestinal systems healthy. The molecule–called absent in melanoma 2–detects the DNA of harmful microorganisms and regulates inflammation in the gut. Cell Nonautonomous Activation of Flavin-Containing Monooxygenase Promotes Longevity and Health Span
Stabilization of the hypoxia-inducible factor 1 (HIF-1) increased life span and health span in nematodes through an unknown mechanism. The authors report that neuronal stabilization of HIF-1 mediates these effects in C. elegans through a cell nonautonomous signal to the intestine, which resulted in activation of the xenobiotic detoxification enzyme flavin-containing monooxygenase-2. Liver X Receptor Regulates Triglyceride Absorption through Intestinal Downregulation of Scavenger Receptor Class B, Type 1
In Caco-2/TC7 cells and mice, intestinal activation of liver X receptor reduced production of chylomicron by a mechanism dependent on the apical localization of scavenger receptor class B, type 1. [Gastroenterology] Abstract

PRMT1-Mediated Methylation of the EGF Receptor Regulates Signaling and Cetuximab Response
Scientists determined that methylation at R198 and R200 of the EGFR extracellular domain by protein arginine methyltransferase 1 (PRMT1) enhances binding to EGF and subsequent receptor dimerization and signaling activation. [J Clin Invest] Full Article | Press Release

MicroRNA-22 Inhibits Tumor Growth and Metastasis in Gastric Cancer by Directly Targeting MMP14 and Snail
Investigators found that the expression of microRNA (miR)-22 was significantly reduced in clinical gastric cancer (GC) tissues compared with paired adjacent normal tissues, and was significantly correlated with a more aggressive phenotype of GC in patients, and low miR-22 expression correlated with poor overall survival. Bile Acids Regulate Intestinal Cell Proliferation by Modulating EGFR and FXR Signaling
Scientists examined the effects of bile acids on intestinal epithelial cell proliferation and investigated the possible roles for epidermal growth factor receptor (EGFR) and farnesoid X receptor (FXR) in these effects. [Am J Physiol Gastrointest Liver Physiol] Abstract

Lgr5 Methylation in Cancer Stem Cell Differentiation and Prognosis-Prediction in Colorectal Cancer
Researchers investigated the methylation status within the leucine-rich-repeat-containing G-protein-coupled receptor 5 (lgr5) promoter and evaluated its relationship with cancer stem cells differentiation, prognosis for colorectal cancer, and its clinicopathological features. Bidirectional Notch Signaling Regulates Drosophila Intestinal Stem Cell Multipotency
Investigators found that during enteroendocrine (ee) cell formation, the ee cell marker Prospero localizes to the basal side of dividing intestinal stem cells. [Science] Abstract

Agonists of the TRAIL Death Receptor DR5 Sensitize Intestinal Stem Cells to Chemotherapy-Induced Cell Death and Trigger Gastrointestinal Toxicity
Researchers showed that the combined administration of the DR5 agonistic monoclonal antibody and chemotherapy to wildtype mice triggered synergistic gastrointestinal toxicities that were associated with the death of Lgr5+ crypt base columnar stem cells in a p53- and DR5-dependent manner.
