Prospective Derivation of a Living Organoid Biobank of Colorectal Cancer Patients
Scientists report the establishment of tumor organoid cultures from 20 consecutive colorectal carcinoma patients. For most, organoids were also generated from adjacent normal tissue. Organoids closely recapitulate several properties of the original tumor. [Cell] Abstract | Graphical Abstract

Reducing DNA Methylation Suppresses Colon Carcinogenesis by Inducing Tumor Cell Differentiation
Scientists found that a genome-wide reduction in the DNA methylation levels induced cellular differentiation in association with decreased cell proliferation in ApcMin/+ mouse colon tumor cells in vivo. [Carcinogenesis] Abstract

An Essential Role of cAMP Response Element-Binding Protein in Epidermal Growth Factor-Mediated Induction of Sodium/Glucose Cotransporter 1 Gene Expression and Intestinal Glucose Uptake

Downregulated Long Noncoding RNA BANCR Promotes the Proliferation of Colorectal Cancer Cells via Downregulation of p21 Expression
Researchers showed that BRAF activated non-coding RNA (BANCR) expression was significantly down-regulated in colorectal cancer tissues compared with normal tissues, and overexpression of BANCR suppressed colorectal cancer cell growth in vitro and in vivo. [PLoS One] Full Article

Commensal Streptococcus salivarius Modulates PPARγ Transcriptional Activity in Human Intestinal Epithelial Cells
Researchers used human intestinal epithelial cell lines engineered to monitor peroxisome proliferator-activated receptor (PPARγ) transcriptional activity in response to a wide range of S. salivarius strains. [PLoS One] Full Article

Altered Expression of Caspases-4 and -5 During Inflammatory Bowel Disease and Colorectal Cancer: Diagnostic and Therapeutic Potential
Scientists demonstrated that caspases-4 and -5 are involved in inflammatory bowel disease-associated intestinal inflammation. They also found robust expression of caspases-4 and -5 within intestinal epithelial cells, exclusively within neoplastic tissue, of colorectal tumours. [Clin Exp Immunol] Abstract

Short Hairpin RNA- Mediated Gene Knockdown of FOXM1 Inhibits the Proliferation and Metastasis of Human Colon Cancer Cells through Reversal of Epithelial-to-Mesenchymal Transformation
Investigators investigated FOXM1 protein expression in 87 colorectal cancer tissue specimens, invasive lymph nodes and adjacent paired normal colorectal tissues by immunohistochemical analysis. [J Exp Clin Cancer Res] Abstract

Mitogen-Activated Protein Kinase Phosphatase-1 Prevents Lipopolysaccharide-Induced Apoptosis in Immature Rat Intestinal Epithelial Cells
The authors demonstrated that mitogen-activated protein kinase phosphatase-1 has a central role in preventing inflammation-induced apoptosis in rIEC-6 enterocytes. [Pediatr Res] Abstract


PERK Limits Drosophila Lifespan by Promoting Intestinal Stem Cell Proliferation in Response to ER Stress

Overlapping DNA Methylation Dynamics in Mouse Intestinal Cell Differentiation and Early Stages of Malignant Progression
Genome-scale analysis of DNA methylation together with microarray gene expression have been applied to compare intestinal crypt stem cells, differentiated cells, ApcMin/+ adenomas and the corresponding non-tumor adjacent tissue, together with small and large intestine samples and the colon cancer cell line CT26.
