Visualization of a Short-Range Wnt Gradient in the Intestinal Stem-Cell Niche
Scientists generated an epitope-tagged, functional Wnt3 knock-in allele. In intestinal organoids, Wnt3-transfer involved direct contact between Paneth cells and stem cells. [Nature] Abstract

Deletion of Men1 and Somatostatin Induces Hypergastrinemia and Gastric Carcinoids
Researchers defined the genetic background that creates a permissive environment for gastric carcinoids using transgenic mouse lines. The combination of conditional multiple endocrine neoplasia 1 gene locus (Men1) deletion in the absence of somatostatin led to the development of gastric carcinoids within two years. [Gut] Abstract

A Network-Based Analysis of Colon Cancer Splicing Changes Reveals a Tumorigenesis-Favoring Regulatory Pathway Emanating from ELK1
Scientists revealed a novel tumor-favoring regulatory pathway involving the induction of the transcription factor MYC by the transcription factor ELK1, and the subsequent induction of the alternative splicing factor PTBP1 by both. [Genome Res] Abstract

Cell Competition Drives the Growth of Intestinal Adenomas in Drosophila
Investigators used the adult fly intestine to investigate the existence and characterize the role of competitive tumor-host interactions. They showed that APC-/-driven intestinal adenomas compete with and kill surrounding cells, causing host tissue attrition. [Curr Biol] Full Article | Graphical Abstract | Press Release

14-3-3 Proteins Regulate Akt Thr308 Phosphorylation in Intestinal Epithelial Cells
The authors report that 14-3-3 proteins control Akt Thr308 phosphorylation during intestinal inflammation. They found that IFNγ and TNFα treatment induce degradation of the phosphoinositide-dependent kinase 1 inhibitor, 14-3-3η, in intestinal epithelial cells. [Cell Death Differ] Abstract

AIM2 Contributes to the Maintenance of Intestinal Integrity via Akt and Protects against Salmonella Mucosal Infection
Researchers demonstrated that Absent in melanoma-2 (AIM2) contributes to the maintenance of intestinal barrier integrity and defense against bacterial infection. [Mucosal Immunol] Abstract

Inhibition of NF-κB Translocation by Curcumin Analogs Induces G0/G1 Arrest and Downregulates Thymidylate Synthase in Colorectal Cancer
Scientists tested the hypothesis that inhibition of NF-κB translocation by curcumin and its analogs EF31 and UBS109 can inhibit cell cycle progression and downregulate thymidylate synthase levels in colorectal cancer cell lines. [Cancer Lett] Abstract

KCTD12 Regulates Colorectal Cancer Cell Stemness through the ERK Pathway
Researchers demonstrated that KCTD12, potassium channel tetramerization domain containing 12, is down-regulated in the cancer stem cell-like cells of colorectal cancer. [Sci Rep] Full Article

Hepatocyte Nuclear Factor 1 Coordinates Multiple Processes in a Model of Intestinal Epithelial Cell Function
The authors investigated the transcriptional network governed by hepatocyte nuclear factor 1 (HNF1) in an intestinal epithelial cell line. Gene ontology process enrichment analysis of the HNF1 targets identified multiple processes with a role in intestinal epithelial cell function, including properties of the cell membrane, cellular response to hormones, and regulation of biosynthetic processes. [Biochim Biophys Acta] Abstract

Selective and Reversible Suppression of Intestinal Stem Cell Differentiation by Pharmacological Inhibition of BET Bromodomains
Investigators demonstrated that mice treated with a small molecule inhibitor of BET bromodomains, CPI203, exhibit greater than 90% decrease in tuft and enteroendocrine cells in both crypts and villi of the small intestine, with no changes observed in goblet or Paneth cells. [Sci Rep] Full Article

Notch Regulation of Gastrointestinal Stem Cells
The authors summarize the literature regarding Notch regulation of gastrointestinal stem cell proliferation and differentiation, highlighting tissue specific functions to compare and contrast Notch in the stomach and intestine. [J Physiol] Abstract
